人肾癌组织EGFR-mRNA原位杂交分析及其意义 被引量:3

In situ hybridization analysis of EGFR mRNA in human renal carcinoma tissues and its significance

摘要 目的 探讨人肾癌组织EGFR mRNA的转录水平、细胞定位及与肾癌生长和发展的关系。 方法 采用Digoxin标记原位杂交法 ,检测 2 5例石蜡包埋存档的肾癌组织和 2 0例相应正常肾组织及配对 16例冰冻保存的肾癌组织和远离癌自身正常肾组织标本的EGFR mRNA转录水平和细胞定位。 结果 肾癌组织和正常肾组织的EGFR mRNA转录阳性率分别为 :石蜡包埋组 44 % (11/2 5 )和 15 % (3/ 2 0 ) ;冰冻保存组 5 0 % (8/ 16 )和 13% (2 / 16 )。两组肾癌组织EGFR mRNA转录阳性率均较正常对照肾组织显著增高 (P均 <0 .0 5 ) ;癌组织原位杂交阳性见于癌细胞 ,主要位于胞核 ,少数位于胞浆 ;而两组正常对照肾组织为弱阳性反应。 结论 人肾癌组织过度转录EGFR mRNA ,可能与肾癌的生长、发展有关。 Objective To study the transcription level of EGFR mRNA in human renal cell carcinoma (RCC) tissues,its cellular location and its relationship with RCC development and growth. Methods In 25 cases of paraffin embedded RCC tissues and 20 cases of corresponding autologous normal kidney ti ssues and in 16 cases of paired RCC tissues and autologous normal kidney tissues frozen and stored -70℃, Digoxin labelled in situ hybridization method was used to analyze the transcription level of EGFR mRNA and its cellular location. Results The positive transcription rates of EGFR mRNA in RCC tissues and normal kidney tissues were respectively 44% and 15% in the paraffin embedded group,50% and 13% in the frozen group.In both groups,there were significantly higher positive transcription rates of EGFR mRNA in RCC tissues than that in normal kidney tissues (P<0.05).The transcribed EGFR mRNA in RCC tissues were seen in the cancer cells,the majority of which located in the nuclei and the minority in the cytoplasms whereas only minimal positive reaction was seen in both groups of normal kidney tissues. Conclusions Overtranscribed EGFR mRNA in human RCC tissues might contribute to the RCC development and growth.
