枯草芽孢杆菌Bs-0728菌株发酵条件的优化 被引量:9

摘要 枯草芽孢杆菌Bs-0728是对苹果再植病害具有良好防治效果的菌株。为了提高Bs-0728发酵液的活菌数量和抑菌效果,本研究通过单因素试验筛选及正交试验,对该菌株进行了液体发酵培养及配方优化。试验中以茄腐镰刀菌FsP4菌株作为指示菌,以抑菌带宽度和OD值作为评价指标,经测试得到最佳培养配方为:玉米粉2%,酵母粉2%,磷酸二氢钠0.1%+磷酸氢二钠0.1%;最适发酵条件为:初始接种量1mL,装液量30mL/300mL,初始pH值7.5,发酵温度32℃,160r/min振荡培养12h。优化后Bs-0728对病原菌的抑菌带宽增加到1.04cm,比初始培养条件下提高了85.7%,OD值达到0.839,比初始培养条件提高了107.7%。 Bacillus subtilis strain Bs-0728 was found to have significant control effects against apple replant disease in vivo and in vitro in previous research in our research group.In order to improve the production and inhibition effect of Bs-0728,the fermentation conditions of Bs-0728 were optimized by using single factor combined with orthogonal experiment design.Fusarium solani isolate FsP4 was selected as an indicating strain,the inhibition zone width and the OD value of culture medium was used as evaluation index.The results showed that the optimal medium was composed of 2%corn meal,2%yeast extract and 0.1% NaH2PO3+0.1% Na2HPO3.The optimal fermentation conditions were that putting 1 mL initial inoculum into 30 mL liquid culture in a 300 mL flask,with the initial pH 7.5 and keeping the fermentation temperature at 32 ℃,160 r/min shaking cultivation for 12 h.Under this optimum condition,the inhibition zone width of Bs-0728 reached 1.04 cm,which was 85.7% higher than that of initial cultural condition;and the OD value of culture medium was 0.839,107.7% higher than that of initial cultural condition.
