摘要
目的建立一种同时测定肾康注射液(大黄、丹参、红花、黄芪)中5种活性成分即芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚方法。方法采用三氯甲烷萃取和RP-HPLC检测,Stamsil-BP C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.2%的磷酸水梯度洗脱,体积流量为1 mL/min,柱温30℃,检测波长为440 nm。结果芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚分别在2.6~130μg/mL(r=0.999 6)、2.67~53.4μg/mL(r=0.999 1)、3.38~67.6μg/mL(r=0.999 3)、6.16~61.6μg/mL(r=0.999 2)、0.32~5.44μg/mL(r=0.999 3)内具有良好的线性关系,加样回收率分别在101.3%、97.9%、103.7%、96.7%及102.4%。结论本方法操作简单、省时、结果准确,重复性好,适用于肾康注射液的质量控制。
AIM To establish an HPLC method for simultaneously determining aloe-emodin, rhein, emodin, chrysophanol, and physcion in Shenkang Injection (Rhei Radix et Rhizoma, Salviae miltiorrhizae Radix et Rhizoma, Carthami Flos, Astragali Radix). METHODS Trichloromethane was used to extract Shenkang Injection and HPLC was performed on Stamsil-BP C18 column (250 mm ×4.6 mm, 5 μm) and the mobile phase was consisted of methanol-0.2% phosphoric acid in gradient elution manner. The flow rate was 1.0 mL/min, the temperature of column was 30 ℃, and the wavelength of detection was set at 440 nm. RESULTS There were good linear rela- tionships for aloe-emodin, rhein, emodin, chrysophanol, and physcion within their concentration ranges of 2.6-130μg/mL (r =0. 999 6), 2.67-53.4 μg/mL (r =0. 999 1), 3.38-67.6 μg/mL (r =0. 999 3), 6. 16-61.6 μg/mL (r=0.9992), 0.32-5.44 μg/mL (r =0.9993) and with good recoveries of 101.3%, 97.9%, 103.7%, 96.7% and 102.4% , respectively. CONCLUSION The established HPLC method is simple and reliable with a good reproducibility, which is suitable for the analysis and quality control of the five anthraquinones.
