摘要
为了阐明野生型p53分热休克蛋白90β(hsp90β)基因表达调控中的作用,我们利用野生型p53的表达质粒转染Jurkat细胞,发现野生型p53可以剂量依赖方式抑制hsp90β基因的启动子活性,并且证实hsp90β基因的表达在mRNA和蛋白水平均明显减少。
Hsp90 acts as a molecular chaperone both constitutively and in cell stress response. Mammollalian Hsp90is encoded by two individual genes, the hsp90α and β isoforms. The two genes are differentially regulated. It hasbeen known that Hsp90 binds to p53 and participates in the regulation of p53 activity. Our results show that wild-typep53 inhibits the expression of hsp90β gene at both mRNA and protein levels in a dose dependent manner. Regulatorymechanisms of p53 on hsp90β gene had been studied and discussed.
