As one importent member of human heat shock proteins,human hsp 90β gene is constitutively experssed in various tissues under physiological condition and it is highly inducible at elevated temperature.A study was carried out to test the methylation status of some CpGs upstream of this gene in Jurkat cells under physiological and heat shock conditions.Extracted genomic DNA from Jurkat cells under both conditions were cut by methylation sensitive restricted endonuclease (Msp Ⅰ/Hpa Ⅱand Hha Ⅰ)and hybridized with human hsp90β cDNA probe.Several CpG dinucleosides sites between -1 102 bp and -173 bp were found non Methylated before and after heat shock treatment.The study indicates that keeping CpG sites upsteam of human hsp90β gene at non methylated status may be needed for the constitutive expression of this gene.
