目的 观察雷公藤内酯醇( TL)对体外培养成骨细胞增殖的影响,以进一步探讨雷公藤致女性骨质疏松的机制。方法 以 Sprague- Dauley(SD)大鼠成骨细胞增殖为模型,分离新生 SD大鼠头盖骨成骨细胞,传代后加入不同浓度 TL。每浓度组设 4个复孔,以 PBS作阴性对照,用 MTT比色分析法检测 TL在体外对成骨细胞增殖的作用和效价。结果 培养 2 d, 10 ng/mL TL显著抑制成骨细胞增殖 (P 【关键词】 雷公藤内酯
Objective The effects of Triptolide( TL) on the proliferation capacity of osteoblasts were studied in order to further explore the mechanism of osteoporosis induced by Tripterygium Wilfordii Hook.F (TW). Methods Osteoblasts were obtained from the calvaria of new- born SD rats and were cultured with various concentrations of TL. The effects of TL on proliferation capacity of osteoblasts were detected by MTT methods. Results The proliferation rates of osteoblasts cultured for 2 days and 4 days were significantly inhibited by 10 ng/mL and 0.01 ng/mL of TL respectively. The inhibition rate was positively correlated with TL concentrations. Conclusion It is indicated that the direct suppressive effects of TL on proliferation capacity of osteoblasts play an important role in the pathogenesis of osteoporosis induced by TW.
