摘要
目的研究外周血造血干细胞简化冷藏保存方法。方法：用ＩＭＤＭ、白蛋白、血浆三种不同保养液于４℃保存造血干细胞，分别在当天、第３至第６天测其存活率、ＣＤ３４阳性率及ＣＦＵ－ＧＭ集落形成能力。结果：造血干细胞保存在４℃以不超过７２小时为好；保养液以自身血浆及４％白蛋白为佳。
Objective: The study aims to find out the simplified method of storage of Peripheral blood stem cells(PBSC). Methods: Used three different kinds of culture liquids- IMDM, 4% albumin and autologous plasma, the PBSC were preserved in 4℃ refrigerator. To assessed the quantity and quality of the PBSC, the viability, CD34 cells and the CFU - GM were tested on day 0, 3, 4, 5, 6 respectively. Results: the PBSC can he preserved in 4 ℃ refrigerator for 72 hours with a good viability and differentiation capacity and autologous plasma,4% albumin are superior to IMDM. Conclusions: PBSC can be preserved in 4 ℃using autologous plasma or 4% albumin, which is an effective methods when the PBSC need not to be preserved for a long Period.
