摘要
目的研究血小板源性生长因子(PDGF)在小肠移植物中的表达及意义。方法利用显微外科技术构建同系和异系大鼠小肠移植模型,以同系移植组作为对照组,异系移植组作为实验组。分别在术后7 d、28 d、90 d进行移植物活检,常规病理学检测,应用Realtime-PCR技术检测移植物内PDGF mRNA转录水平,采用免疫组织化学方法对PDGF的表达进行定位。结果检测显示,所有移植物中PDGF mRNA转录水平总体呈上升趋势,到实验终点即POD90所采集的标本中以实验组的mRNA转录水平最高。免疫组化可见,PDGF在间质及小肠腺体内高表达。结论 PDGF因子在终末期小肠移植物中高表达,说明PDGF因子对于小肠移植慢性排斥反应有重要提示意义。
Objective To analyse the expression of PDGF in small bowel transplantation in rats.Methods Isogeneic and allogeneic small bowel transplantation were performed in rats with microsurgical technology.Recipients were divided into two groups： isogeneic control group and allogeneic test group.Graft tissues were harvested on POD7,POD28 and POD90.mRNA transcription and immunohistochemical staining were also performed to detect the expression of PDGF.Results The expression of PDGF showed ascending tendency in whole stage,and the results of test group of POD90 were significantly higher than that of other stages.The immunohistochemical staining showed a higher expression in graft tissue in test group.Conclusion PDGF is highly expressed in the end stage of small bowel transplantation,and shows meaningful use for examination of chronic rejection.
