摘要
为探讨转化生长因子β_1(TGFβ_1)在人体血液动力学超负荷性心肌肥厚发生机制中的作用,用天狼猩红特染、免疫组织化学及原位杂交技术检测了32例(左、右室心肌各16例)肥大心肌组织中胶原、TGFβ_l mR-NA和蛋白的表达情况。结果显示,压力和容量超负荷都可使心肌组织胶原增生,TGFβ_1mRNA和蛋白表达水平显著增加。 TGFβ_1 mRNA和蛋白表达水平与心肌细胞横径增加呈显著正相关, TGFβ_1 mRNA和蛋白表达水平与心肌胶原总量也呈显著正相关。说明TGFβ_1表达可能是介导人体超负荷性心肌肥厚发生的因素之一。
To investigate the role of TGFβ_1 in the process of human heart hypertrophy induced by overload. Sirius Red staining, immunochemistry and in situ hybridization were used to detect collagen, TGFβ_1, gene expression in 32 human heart hypertrophy(right and left ventricle, each 16). The results showed that total collagen, TGF6, mRNA and protein expression were elevated significantly in hypertrophic myocardium (P<0. 05-0. 01), and the rate of collagen Ⅰ and Ⅲ was also increased as compared with control. TGFβ_1, mRNA and protein expres- sion level were positively related with left and right myocardial hypertrophy, and TGFβ_1, mRNA and protein ex- pression were also positively corelated with total collagen of hypertrophic myocardium. TGFβ_1, mRNA and protein expression may play a role in the mechanism of human heart hypertrophy induced by overload.
