摘要
目的 :探讨cyclinD1蛋白与霍奇金淋巴瘤 (HL)R S/H细胞凋亡及增殖的关系。 方法 :利用免疫组化检测了 40例HL中R S/H细胞bcl 2、fas及cyclinD1的表达。结果 :bcl 2和fas蛋白阳性信号在R S/H细胞中主要呈浆 膜型。在HL 4种亚型 (LP、NS、MC和LD)中bcl 2阳性细胞密度逐渐增高 (P <0 0 5 ) ,而fas阳性细胞密度依次降低 (P <0 0 1) ,并呈明显负相关(r =- 0 5 6 90 )。在 40例HL中cyclinD1阳性表达率为 70 % (2 8/ 40 ) ,阳性信号主要位于核内 ;cyclinD1阳性病例中的bcl 2阳性细胞密度显著高于fas阳性细胞密度 (P <0 0 1) ,而在cyclinD1阴性病例中两者差异无显著性 (P >0 0 5 ) ;在cyclinD1阳性和阴性病例中 ,bcl 2和fas阳性细胞密度均呈明显负相关 (分别r =- 0 6 131,r =- 0 7310 )。结论 :cyclinD1可能在HLR S/H细胞增殖中与bcl 2蛋白具有协同作用 ,并与fas蛋白一起调控R S/H细胞的增生与凋亡 ;bcl 2和fas的表达与HL不同组织学亚型及预后有关。
Purpose To study the relationships between the expressions of cyclin D1 protein, bcl 2 and fas protein and apoptosis and proliferation of R S/H cells in Hodgkins lymphoma (HL). Methods Immunohistochemical technique S P method was used to detect expressions of bcl 2, fas and cyclin D1 proteins of R S/H cells in 40 cases of HL. Results The positive signals of bcl 2 and fas protein were mainly cytoplasm membrane type in R S/H cells. The density of positive cells for bcl 2 protein increased gradually (P<0 05), and for fas decreased gradually (P<0 01). Both densities were correlated negatively in linear fashion (r=-0 5690) among subtypes of HL (LP, NS, MC and LD). The positivity rate of cyclin D1 protein expression in 40 cases of HL was 70% (28/40). The positive signal of cyclin D1 was mainly localized in nuclei of R S/H cells. In the cases for cyclin D1 protein, the density of bcl 2 positive cells was significantly higher than that of fas positive cells (P<0 01), and the difference of bcl 2 expression to fas expression was not found in the cases for cyclin D1 protein negative (P>0 05). There was a negative relationship between bcl 2 and fas positive cell densities whether in the cases for cyclin D1 protein positive (r=-0 6131) or for cyclin D1 protein negative (r=-0 7310). Conclusion It suggest that cyclin D1 protein may cooperate with bcl 2 protein in proliferation of R S/H cells, and regulate proliferation and apoptosis of R S/H cells with fas protein together. The expression of bcl 2 and fas proteins may be associated with the subtype and prognosis of HL.
