摘要
该文利用免疫组化检测了40 例霍奇金病( HD) 中RS/ H 细胞bcl- 2 、fas 及cyclin D1 的表达,并对其相关关系进行了研究。结果显示:bcl - 2 和fas 蛋白阳性信号在RS/ H 细胞中主要呈浆膜型。在HD 四种亚型LP,NS,MC 和LD 中bcl- 2 阳性细胞密度逐渐增高( P< 0 .05) ,而fas 阳性细胞密度依次降低( P< 0 .05) ;并呈明显负相关(r = - 0 .5690) 。在40 例HD 中cyclin D1 阳性表达率为70 % (28/40) ,阳性信号主要位于核内;cyclin D1 阳性病例中的bcl- 2 阳性细胞密度显著高于fas 阳性细胞密度( P< 0 .01) ,而cyclin D1 阴性病例中两者无显著差异( P<0 .05) ;在cyclin D1 阳性和阴性病例中,bcl - 2 和fas 阳性细胞密度均呈明显负相关( 分别r = - 0 .6131 ,r =- 0 .7310) 。提示抗凋亡基因bcl- 2 和促凋亡基因fas 的表达与HD 不同组织学亚型及预后有关;cyclin D1 蛋白可能具有促进bcl - 2 的表达和/ 或抑制fas 表达作用,从而导致RS/ H
The expressions of bcl-2,fas and cyclin D1 protein were detected by immunohistochemistry in RS/H cells of 40 cases of Hodgkin's disease(HD),and their correlationshipsn were analyzed.The results showed that the positive expressions of bcl-2 and fas proteins were mainly cytoplasm-membrane type in RS/H cells.The density of positive cells for bcl-2 protein increased gradually(P<0.05),while the density of fas protein decreased gradually(P<0.01); both densities were negatively correlated in linear fashion(r=-0.5690) in the subtypes of HD(LP,NS,MC and LD).The positive rate of cyclin D1 was protein expression in 40 cases of HD was 70%(28/40).The positive expression of cyclin D1 was located in the nuclei of the RS/H cells.In the positive cases of cyclin D1 protein, the density of bcl-2 positive cells was significantly higher than that of the fas positive cells (P<0.01);a correlation between bcl-2 expression and fas expression was found in the cases of negative cyclin D1 protein (P<0.05).There was a negative correlation between the densities of bcl-2 and fas positive cells both in the cases of positive cyclin D1 protein(r= 0.6131)and negative cyclin D1 protein(r=-0.7310).The results suggest that the expression of bcl-2 and fas proteins may be associated with the subtype and prognosis of HD;and cyclin D1 overexpression may result in the imbalance of the proliferation and apoptosis of RS/H cells by promoting the expression of bcl-2 protein and/or reducing the expression of fas.
