摘要
目的:分析并比较卵巢癌细胞中与肿瘤转移、侵袭相关的转录因子。方法:采用细胞划痕及小室跨膜实验比较卵巢癌细胞HO-8910及其高转移株HO-8910PM的转移侵袭能力差异;采用高通量转录因子活性检测芯片(DNA/蛋白结合芯片)分析并比较两种细胞中转录因子的活化情况,明确与肿瘤侵袭、转移相关表达的转录因子;用转录因子酶联免疫活性分析方法验证芯片检测结果。结果:细胞划痕实验及侵袭实验表明HO-8910PM比HO-8910细胞转移侵袭能力强,在345个转录因子中共筛选出与肿瘤侵袭、转移相关的31个表达差异转录因子。HO-8910PM细胞与HO-8910细胞相比,表达上调的有13种,下调的有18种。v-Maf仅表达于HO-8910PM,HLF仅表达于HO-8910。结论:v-Maf、HLF等31种转录因子可能参与了卵巢癌侵袭转移的相关机制。
Objective:To analyze the potential transcriptional factors associated with ovarian cancer invasion and metastasis.Method:Ovarian cancer cells HO-8910 and HO-8910PM were used as a pair of metastasis in vivo cellular model.The cellular invasion and metastasis ability of HO-8910 and HO-8910PM were compared by scarification assay and millicell chamber artificial reconstituted basement membrane invasion assay.Furthermore,345-channel Protein/DNA array was used to analyze the potential transcriptional factors with invasion and metastasis.Results:A total of 345 candidate transcriptional factors,31 transcriptional factors were associated with migration and metastasis.Compared with HO-8910,31 transcriptional factors showed a different activies in HO-8910PM cells,which including 13 up-regulation and 18 down-regulation.Conclusion:DNA-protein microarry is a useful method to screen the potential transcriptional factors associated with ovarian cancer invasion and metastasis.Transcriptional factors such as v-Maf 、HLF may play an important role in ovarian cancer.
