摘要
目的:观察叶酸对胚胎大鼠心肌细胞Cx43表达的影响,探讨叶酸预防先天性心脏畸形作用的可能机制。方法:受孕大鼠随机分为两组,实验组给予叶酸(2mg/kg/d)灌胃,对照组灌服等量生理盐水。选择ED13.5和ED16.5的胚胎大鼠作为研究对象,SP免疫组织化学染色法观察Cx43在心肌细胞的表达。结果:免疫组织化学染色显示各组胎鼠心肌细胞均可见Cx43阳性表达,阳性产物以颗粒形式散在分布于细胞质内,或呈短线状、链状排列在细胞侧-侧连接处。实验组胎鼠心肌Cx43阳性细胞数明显高于同胎龄对照组(P<0.01)。结论:叶酸可增强胎鼠心肌细胞Cx43的阳性表达。
Objective:To observe the effects of folic acid on expression of Cx43 in myocardial cells of embryonic rat,and investigate the possible mechanisms of folic acid preventing congenital heart disease.Methods:Pregnant rats were randomly divided into 2 groups: the rats in experimental group were lavaged with folic acid(2mg/kg/d),while rats in control group were lavaged with equivalent physiological saline.SP immunohistochemical staining was used to observe the expression of Cx43 in myocardial cells of ED13.5 and ED16.5 embryonic rats.Results:The Cx43 immunopositive products distributed in cytoplasm of myocardial cells,or located at cell side-side joint in spot-band or chain-shaped.The Cx43 positive myocardial cell number of embryonic rats in experimental group were obviously higher than that of embryonic rats in control group at the same gestational age(P0.01).Conclusions:Folic acid can increase positive expression of Cx43 in myocardial cells of embryonic rats.
