摘要
目的:建立测定白花蛇舌草配方颗粒中槲皮素和山奈素含量的HPLC方法。方法:采用正交试验优化白花蛇舌草配方颗粒的提取工艺,用HPLC测定白花蛇舌配方颗粒中槲皮素和山奈素的含量。色谱柱为Hypersil ODS C18柱(4.6 mm×250 mm,5μm),流动相甲醇-0.2%磷酸(45∶55),检测波长360 nm,流速1.0 mL.min-1,柱温30℃。结果:槲皮素在0.09~0.72μg线性关系良好(r=0.999 9),山奈素在0.034 3~0.274 4μg线性关系良好(r=0.999 9),槲皮素、山奈素的平均加样回收率分别为100.80%(RSD 1.79%),101.75%(RSD 1.76%)。结论:该方法简便、快速、重复性好,可作为白花蛇舌草配方颗粒中槲皮素和山奈素的含量测定。
Objective: To establish an HPLC method for determination of quercetin and kaempferol in Baihua Sheshecao dispensing granule. Method: Orthogonal experiments were used to optimize extraction technology. The samples were separated on a Hypersil ODS C18 (4.6 mm× 250 mm, 5 μm) column which was eluted with methanol-0. 2% phosphoric acid solution (45:55) with detective wavelength at 360 nm and flow rate at 1 mL ·min^-1, column temperature at 30 ℃. Result: The method had good linear relationship within the range of O. 09-0.72 i^g (r = 0. 999 9) for quercetin and 0. 034 3-0. 274 4 ixg ( r = 0. 999 9) for kaempferol. The average recovery rate were 100.80% (RSD 1.79% ) and 101.75% (RSD 1.76% ). Conclusion: The method is simple, rapid and reliable which can be used to determine quercetin and kaempferol in Baihuashe Shecao dispensing granule.
