摘要
目的:构建ABCG2过表达慢病毒载体,并在人乳腺癌细胞微球体细胞(mammospheres-derived cells,MSDCs)中建立稳定表达模型。方法:应用ABCG2过表达慢病毒载体转染由无血清悬浮培养的MSDCs,并计算其转染效率。结果:成功构建ABCG2过表达慢病毒载体,在MSDCs中其转染效率为92.4±1.8%。结论:ABCG2过表达慢病毒载体能在MSDCs中稳定表达。
Objective: To construct a lentiviral overexptession vector carrying ABCG2, and establish a stable experession model of mammospheres - derived cells (MSDCs). Methods: The lentiviral over expression vector carrying ABCG2 was transduced into MSDCs, and the transduction efficiency was calculated. Results:The lentiviral over expression vector carrying ABCG2 was constructed suecesdully, and the transduction efficiency was 92.4 ± 1.8 %. Canclusion:The lentiviral over expression vector carrying ABCG2 can be expressed stably in MSDCs.
