原发性高血压与血管紧张素转换酶基因多态性的相关性研究 被引量:6

摘要 血管紧张素转换酶 (ACE)作为肾素—血管紧张素系统的一个关键酶 ,可能参与高血压的发病。ACE水平由遗传决定 ,有三种基因型 :纯合子Ⅱ、DD型和杂合子ID型。本研究探讨ACE基因多态性与原发性高血压(EH)的关系。研究人群为 10 4例EH患者及 113例正常人。ACE基因多态性检测用聚合酶链反应 (PCR) ,同时用比色法测定其血清ACE水平。结果发现 :①ACE基因多态性与EH明显相关 ,EH者DD基因型明显高于正常人 ;②ACE基因多态性与血清ACE水平明显相关 (DD >ID ,DD >Ⅱ )。本研究提示ACE基因DD型与EH有关 ,并与血清ACE水平一致 。 WT5”BZ]Angiotensin converting enzyme(ACE)which is a key enzyme in the renin angiotensin system has been suggested to be involved in the development of essential hypertension(EH).The ACE levels is determined genetically.The ACE gene is divided into three genotypes(Ⅱ and DD homozygotes,and ID heterozygotes).In the present study,we examined the relationship between the genotypes of the ACE gene and EH.The study population consisted of 104 hypertensive and 113 normal subjects.The genotypes of the ACE gene was determined by the polymerase chain reaction(PCR).The serum ACE concentration was measured by high performance liquid chromatography with the use of an artificial substrate.The results were as following:①A significant relationship has been found between the polymorphism of the ACE gene and EH,the DD genotype seems to predispose to LVH.②The ACE gene I/D polymorphism is strongly associated with serum ACE levels(DD>ID,DD>Ⅱ).Findings imply that EH is partially determined by genetic disposition(the polymorphism of ACE gene),and consistent with the increasing serum ACE levels.Furthermore study about the mechanism is necessary.
