摘要
对单纯用二乙基亚硝胺 (DENA)及氯化钆 (GC)或酵母多糖 (ZM)分别阻断或激活 Kupffer细胞后 ,同时给以 DENA所引起的大鼠肝癌中的增殖细胞核抗原 (PCNA )、 bax、 p5 3蛋白的表达和肝癌细胞的凋亡运用免疫组织化学和 TU NEL 法进行了对比研究 ,以探讨 Kupffer细胞在大鼠实验性肝癌细胞凋亡中的作用。结果显示 :肝癌组织的增殖指数、凋亡指数在 C组、 A组、 B组分别呈增高或降低趋势 ;bax、 p5 3阳性率在 C组明显高于 A组 (P<0 .0 5 ) ,同时 ,bax、p5 3蛋白的表达与凋亡指数存在正相关。结果提示 ,Kupffer细胞可能促进实验性肝癌细胞凋亡。
To explore the influence of Kupffer cells on apoptosis in the experimental hepatocellular carcinoma (HCC), a comparative study on the expression of proliferating cell nuclear antigen (PCNA), bax, P53 and apoptosis of liver cancer cells was performed by using immunohistochemistry and terminal deoxynucleotydil transferase (TdT) mediated dUTP digoxigenin nick end labeling (TUNEL) in diethylnitrosamine (DENA) induced HCC in rats with and without pretreatment with DENA (group A) or gadolinium chloride (GC, group B) or zymosan (ZM, group C) which might respectively block or enhance the activity of Kupffer cells. The results showed the proliferating index (PI) was increased and apoptosis index (AI) decreased in group C, group A and group B. The positive rate of bax and P53 in group C was significantly higher than in group A ( P <0.05). There was a positive correlation between the AI and expression of both bax and p53 protein. The results suggested that Kupffer cells might promote apoptosis in the experimental HCC.
