子宫颈癌多药耐药基因的表达及其临床意义 被引量:6

摘要 目的 探讨多药耐药基因 (MDR1)在子宫颈癌组织中的表达及其临床意义。方法 利用逆转录聚合酶链反应 (RT -PCR)方法检测 46例子宫颈癌组织MDR1基因表达情况 ,同时利用四甲基偶氮唑篮比色法(MTT)检测肿瘤细胞对化疗药的敏感性。结果  2 8例 (6 1% )宫颈癌组织MDR1基因表达阳性 ,MDR1基因表达与肿瘤细胞组织学分级有关 (P <0 0 5 ) ,而与肿瘤组织学类型及临床分期无关。MDR1表达阳性的宫颈癌细胞对长春新碱、阿霉素和足叶乙甙交叉耐药。结论 MDR1基因表达阳性的子宫颈癌可能对长春新碱、阿霉素和足叶乙甙耐药 ,顺铂可作为子宫颈癌化疗的首选药物。 Objective To study the expression and clinical significance of multidrug resistance gene(MDR1)in cervical cancer.Methods In 46 untreated patients with cervical cancer,the expression of MDR1 gene was detected using retrotranscription PCR(RT-RCR).Sensitivity to anticancer agents was also examined by MTT assay.Results MDR1 gene was positive in 28(61%)cervical cancers.MDR1 gene expression was significantly correlated with the differential grade of tumor cells,but there was no confirmed relationship between MDR1 gene expression and the pathologic type or stage.Cervical cancer cells with positive MDR1 gene expression showed cross drug resistance to vincristine(VCR),adriamycin(ADM),and etoposide(VP16).Conclusion Cervical cancer with MDR1 gene positive expression may be resistant to VCR,ADM and VP16.Cisplatin could be the first choice for initial therapy of cervical cancer.The expression of MDR1 gene may be related to the prognosis of cervical cancer.
