摘要
目的探讨子痫前期及子痫患者β-纤维蛋白原(Fg)-455G/A基因多态性的分布及其与血浆Fg浓度、子痫前期及子痫发生和发展的相关性。方法应用聚合酶链反应和限制性内切酶片段多态性技术(PCR-RFLP)检测2010年9月至2011年6月第三军医大学大坪医院野战外科研究所收治的56例正常妊娠妇女及50例子痫前期、子痫患者的β-Fg-455G/A基因多态性,凝固法测定血浆Fg水平。结果正常妊娠组与子痫前期及子痫组比较,GG、GA、AA基因型分布频率差异无统计学意义,A基因频率之间、G基因频率之间差异也无统计学意义。但轻度子痫前期与重度子痫前期、子痫比较,重度子痫前期及子痫患者中A等位基因携带者(GA+AA)明显多于轻度子痫前期患者(P<0.05)。无论正常妊娠组还是子痫前期及子痫组,A等位基因携带者血浆Fg水平均显著高于GG基因型者。子痫前期及子痫组中,GA+AA基因型收缩压及舒张压均明显高于GG型。结论β-Fg-455G/A基因多态性与子痫前期及子痫发病及病情轻重程度有一定相关性。
Objective To explore the distribution of β-Fibrinogen-455G/A gene polymorphism and the association of the polymorphisms with concentration of Fg and preeclampsia and eclampsia.Methods The β-Fg-455G/A gene polymorphism was identified by polymerase chain reaction(PCR)and restriction fragment length polymorphism(RFLP) in 56 normal pregnancy women and 50 patients with preeclampsia and eclampsia.Turbidimetric assays were performed to measure the plasma fibrinogen levels.Results Compared with normal pregnancy,genotype(GG,GA,AA) frequency and alleles(G,A) frequency of preeclampsia and eclampsia patiens were no obviously changed(P0.05).Compared with mild preeclampsia,severe preeclampsia and eclampsia have more A-allelic(GA+AA)(P0.05).The A-allelic is associated with elevated plasma fibrinogen levels in both preeclampsia and eclampsia and controls.In preeclampsia and eclampsia group,both systolic pressure and diastolic pressure of the A-allelic(GA+AA) were obviously higher than the genotype of GG(P0.05).Conclusions Although our study haven't find the association between β-Fg-455A allele gene and the morbidity of preeclampsia and eclampsia,it may correlate to severity of preeclampsia and eclampsia.
