6种开心散类方对不同物质损伤神经细胞的保护作用 被引量:17

Protective effect of six Kaixin San formulas on nerve cells injured by different materials

摘要 目的:研究组方相同,比例不同的6种开心散类方对模拟抑郁、阿尔茨海默病和帕金森病细胞模型的保护作用。方法:以0.4 mmol·L-1皮质酮损伤SH-SY5Y细胞,20μmol·L-1β-淀粉样肽(Aβ25-35)损伤PC12细胞,250μmol·L-11-甲基-4-苯基吡啶离子(1-methyl-4-phenylpyridinium,MPP+)损伤SH-SY5Y细胞,建立体外模拟抑郁、阿尔茨海默病及帕金森模型,采用MTT法检测细胞的存活率及LDH法检测细胞损伤程度。结果:100,500 mg·L-1《备急千金要方》之定志小丸可显著提高皮质酮损伤的SH-SY5Y细胞的存活率并明显减少LDH的释放浓度;各开心散类方均有明显增加Aβ25-35损伤的PC12细胞的存活率、减少LDH的释放浓度的作用;其中以10,100,500 mg·L-1《备急千金要方》之开心散的作用最为显著;500 mg·L-1《古今录验》之茯神丸可显著提高MPP+损伤的SH-SY5Y细胞的存活率、减少LDH的释放。结论:定志小丸对皮质酮损伤的神经细胞有明显的保护作用,有潜在抗抑郁作用;6种开心散类方对Aβ25-35损伤的PC12细胞均有保护作用,有潜在抗阿尔茨海默病作用,《备急千金要方》之开心散作用最为显著;《古今录验》之茯神丸高剂量组对MPP+损伤的SH-SY5Y细胞有一定的保护作用。 Objective: To investigate the protective effect of six Kaixin San formulas on simulated cells model of depression, Alzheimer's disease and Parkinson's disease. Method: The in vitro simulated cells model of depression, Alzheimer's disease and Parkinson's disease was established by injuring SH-SYSY cells with corticosterone (0.4 mmol ·L^-1 ) , injuring PC12 cells with neurotox- ic amyloid peptide (Aβ25-35) (20 μ mol·L^-1) and injuring SH-SYSY cells with 1-methyl-4-phenylpyridinium ion (MPP + ) (250 μ mol·L^-1). The cell survival rate was assayed with MTY method and the degree of cell injury was detected with LDH. Result: 100, 500 mg·L^-1 Dingzhixiao Wan prepared as mentioned in Beiji Qianjin Yaofang could significantly increase the survival ratio of SH-SY5Y cells injured by corticosterone and reduce LDH concentration released. All of the Kaixin San formulas could significantly increase the survival ratio of PC12 cells injured by Aβ25-35 and reduce LDH concentration released. Particularly, Kaixin San( 10, 100, 500 mg·L^-1) prepared as mentioned in Beiji Qianjin Yaofang shown the best effect. And 500 mg·L^-1 Fushen Wan prepared as mentioned in Gujin Luyan could significantly increase survival ratio of SH-SY5Y cell injured by MPP + and reduce LDH concentration released. Conclusion: Dingzhixiao Wan prepared as mentioned in Beiji Qianjin Yaofang could protect corticosterone-induced SH-SYSYcells injury, showing a potential antidepressant effect. All of the six Kaixin San formulas could protect Aβ25-35- induced PC12 ceils injury, but Kaix- in San prepared as mentioned in Beiji Qianjin Yaofang shown the best potential effect for Alzheimer's disease. Fushen Wan prepared as mentioned in Gujin Luyan could protect MPP + -induced SH-SY5Y cells injury to some extent.
