草莓上胶孢炭疽菌的分离与鉴定 被引量:13

Isolation and Identification of Pathogenic Fungus Colletotrichum gloeosporioides(Penz. ) Sacc. Causing Strawberry Anthracnose

摘要 从田间感染炭疽病的草莓组织获得分离物,对其进行培养与纯化,并进行形态学观察,初步鉴定为胶孢炭疽菌(Colletotri-chum gloeosporioides(Penz.)Sacc.),同时利用通用引物ITS1和ITS4对该病原菌rDNA的ITS序列进行PCR扩增,经聚类分析,鉴定结果与形态学鉴定一致,即分离的病原菌为胶孢炭疽菌。采用人工喷雾接种法将胶孢炭疽菌接种到‘章姬’和‘阿尔比’2个栽培草莓品种以及野生黄毛草莓上,3个检测对象对该致病菌株的抵抗力存在一定的差异:‘章姬’为高度感病,‘阿尔比’为感病,黄毛草莓为中等抗病。 Strawberry anthracnose disease tissues were obtained in the field,in order to separate and purify pathogens.Morphology identification demonstrated that the pathogens were Colletotrichum gloeosporioides(Penz.) Sacc..PCR amplifications were performed for the ITS regions of rDNA using the universal primers ITS1 and ITS4.The ITS sequences were deposited in GenBank and compared against those sequences already found in the databases using the BLAST search.Phylogenetic analysis indicated that the pathogens isolated from disease strawberry plants were Colletotrichum gloeosporioides,that was consistent with the identification of morphological characteristics.Colletotrichum gloeosporioides spore suspension were inoculated in cultivars strawberrys 'Akihime','Albion' and a wild species Fragaria nilgerrensis Schlecht.using spraying inoculation method.Results showed that the strain was able to pathogenic.The resistance of three samples were different: 'Akihime' was a high susceptible cultivar,'Albion' was susceptible cultivar and F.nilgerrensis Schlecht.was moderately resistant.
