摘要
目的探讨辛伐他汀对支气管哮喘(简称哮喘)模型大鼠血清中白介素4(interleukin-4,IL-4)和干扰素-γ(interferon-γ,IFN-γ)含量变化的影响,为辛伐他汀治疗哮喘提供实验依据。方法选取SD雄性大鼠32只,随机分为4组:正常对照组、哮喘模型组、地塞米松治疗组和辛伐他汀治疗组。每组各8只,采用卵白蛋白(ovalbumin,0VA)致敏和雾化吸人激发制备大鼠哮喘模型,用双位点夹心酶联免疫吸附试验测定各组大鼠血清中IL-4、IFN-γ含量。结果哮喘模型组血清中IFNγ含量为(83.71±8.89)ng/L明显低于正常对照组;1174含量为(106.62±13.33)ng/L,明显高于正常对照组,差异有统计学意义(P〈0.01)。与哮喘模型组相比,辛伐他汀组和地塞米松组血清中IFN-γ含量分别为(95.93±11.74)ng/L和(96.34±11.64)ng/L,明显高于哮喘模型组,差异有统计学意义(P〈0.05),而两个治疗药物组之间差异无统计学意义(P〉0.05)。与哮喘模型组相比,两个治疗药物组的IL-4含量分别为(79.08±7.34)ng/L和(73.39±7.76)ng/L,均有明显降低,差异有统计学意义(P〈O.01),但两个治疗药物组间差异无统计学意义(P〉0.05)。结论辛伐他汀和地塞米松均能显著提高哮喘大鼠血清IFN-γ的含量,降低血清IL-4含量。
Objective To study the impact of simvastatin on the asthma model rats serum interleukin-4(IL-4) and interferon-γ(IFN-γ) level changes, which provide the experimental basis for simvastatin treatment of bronchial asthma. Methods 32 male SD rats were randomly divided into four groups:normal control group, asthmatic model group, the dexamethasone treatment group and the simvastatin group,with 8 rats in each group, using ovalbumin (OVA) sensitization and aerosol inhalation to stimulate the rat model of asthma. Double sites sandwich enzyme-linked immunosorbent assay in serum IL-4 and IFN-γ levels. Results The levels of serum IFN-γ were significantly lower than the normal control group, IL-4 content was significantly higher than the normal control group, has a very significant difference ( P〈0.01), respectively (83.71 ±8.89) ng/L and (106.62 ± 13.33) ng/L; IFN-γ in serum compared with asthmatic model group, sirnvastatin groupand dexamethasone group were (95.93±11.74) ng/L and (96.34 ± 11.64) ng/L, significantly higher than the asthma model group, a significant difference (P 〈0.05), and no significant difference (P 〉 0.05) between the two treatment groups; compared with asthmatic model group, IL-4 levels in both treatment groups were significantly lower ( P 〈0.01) of IL-4 levels were (79.08 ± 7.34) ng/L and (73.39 ±7.76) ng/L in no significant difference between therapy groups ( P 〉0.05). Conclusions Simvastatin and dexam-ethasone can significantly improve the level of serum IFN-γ in asthmatic rats, reducing the level of serum IL-4 had better control of asthma.
