摘要
目的:分别研究丹参的醇溶性成分和水溶性成份,以得到纯度较高、得膏率较低的浸膏,提高收率,便于制剂成型。方法:采用正交试验法分别优选出丹参醇溶性有效成分和水溶性有效成分的最佳提取工艺,同时考察了提取次数对提取率的影响,并用验证实验证明了该工艺的可行性;采用大孔树脂层析法精制丹参水溶性成分,通过树脂型号的选择、上样液浓度的考察、洗脱剂的优选等层析条件的考察确定了丹参精制的工艺条件。结果:确定丹参的提取精制工艺:丹参药材用6倍量95%乙醇加热回流,提取1h,减压回收溶剂,得稠浸膏减压干燥,粉碎,即得丹参醇溶性提取物;将丹参醇提后药渣用8倍量水煎煮1h,提取液滤过,滤液减压浓缩至约1.5g生药/mL,离心后取上清液作为上样液,通过AB-8大孔树脂至饱和。上样结束后,用去离子水洗脱,至Molish反应呈阴性,停止洗脱。换70%乙醇为洗脱液洗脱,至洗脱液的三氯化铁-铁氰化钾试液反应呈阴性,合并洗脱液,减压回收溶剂,减压干燥,粉碎,即得丹参水溶性提取物。结论:优选的提取精制工艺,可提高药材中有效成分的收率,降低得膏率,便于制剂成型。
Objective: To study water- soluble and alcohol -soluble components in Salviae Miltiorrhizae Radix so to obtain extracts with high purity and low rate of the solid products, and to enhance the yield and facilitate the preparation molding. Methods : The optimized conditions of 'alcohol - soluble and water - soluble active ingredients were selected by orthogonal tests, after which the times of extractions were investigated. And the verification experiments were carried out to prove the feasibility of the processes. Macroporous - resin adsorption technology was applied with the selection of the type of resin, the concentration of the extraction liquid and the washing agent to determine the refining conditions of water - soluble components. Results :The determined preparation process of extracts of Salviae Miltiorrhizae Radix was as the following. The herb was extracted by six - fold ethanol for one hour, after which the extracts were concentrated and dried with vacuum conditions. Then the alcohol - soluble components were obtained. The residue of the alcohol extractions were extracted by eight - fold water for another hour. The water - extracted liquid was concentrated to about 1.5g herb/mL as the loading sample after centrifugation, which were added to AB - 8 macroporous - resin column to refine until the positive outflow were detected. And then water was applied to wash the column until Molish reaction showed negative. 70% etha- nol was used as the washing agent until no positive outflow was detected. The water - soluble extracts were obtained after the washing outflow was concentrated and dried with vacuum. Conclusions: The selected extracting and refining process increased the yield of active ingredients in Salviae Mihiorrhizae Radix, reduced the solid rates, which were favorable to the preparation molding process.
