摘要
目的:观察右归饮协同骨髓干细胞(MSCs)移植对液氮冷冻股骨头坏死家兔模型骨修复的影响。方法:40只兔随机分为4组:A组(模型组)、B组(右归饮组)、C组(干细胞移植组)、D组(右归饮加干细胞移植组)。分离、培养、增殖MSCs;改良液氮冷冻法造模成功1周后,B组采用右归饮灌胃治疗,C组行髓芯减压术后MSCs 1mL(细胞密度:1×105·mL-1)移植治疗,D组采用右归饮协同MSCs移植治疗,对一般形态学、4、7周后的股骨头组织病理学和RT-PCR观察VEGF表达情况进行检测。结果:各治疗组较A组的观测指标均有明显改善,其中各治疗组空骨陷窝阳性率和VEGF阳性表达与A组组间比较差异均有统计学意义(P<0.01),且D组改善优于B、C两组(P<0.05)。结论:右归饮协同骨髓干细胞(MSCs)移植法能够显著改善冷冻坏死股骨头的血供、增强VEGF表达,促进坏死骨修复。
Objective: To observe the effect of Youguiyin combined with Mesenchymal Stem Cells (MSCs) Transplanta- tion for repairing rabbit model osteonecrosis of femoral head induced by liquid nitrogen refrigeration. Methods :40 rabbits were randomly divided into 4 groups:group A( model group), group B (Youguiyin group) , group C (MSCs transplantation group), Group D (Youguiyin with MSCs transplantation group). MSCs were isolated, eultureed, and proliferated. After one week of improved liquid nitrogen freezing method modeling, rabbits in group B were intragastrieally administrated with Youguiyin, rabbits in group C were administrated with 1 mL of MSCs ( cell density : 1×10^5/mL) transplantation after coredecompression ; rabbits in group D were administrated with Youguiyin combined with MSCs transplantation. 4 and 7 weeks after treatment,the general morphology of rabbits and histopathology of the femoral head were observed, and the expres- sions of VEGF were evaluated by RT - PCR. Results -The observations of treatment groups were significantly improved than that in group A. The differences of empty lacunae positive rate and positive expression of VEGF in each treatment group and group A were statistically significant ( P 〈 0.01 ), and Group D improved better than group B or C ( P 〈0. 05 ). Conclu- sion : The combination of Youguiyin with transplantation of MSCs has significant therapeutic effects on the the freezing nec- rosis of the femoral head, can significantly improve blood supply, enhance expression of VEGF to promote the repair of necrotic bone.
