摘要
[目的]探讨不同抗生素对矮牵牛叶片再生的影响。[方法]以开花前顶部新展开的矮牵牛叶片为外植体,接种于添加不同抗生素(卡那霉素、头孢唑林钠和潮霉素)的基本培养基MS+2.0μg/ml BA+0.2μg/ml NAA上,研究不同抗生素对叶片再生的影响。[结果]20 mg/L卡那霉素、5 mg/L潮霉素可明显抑制矮牵牛叶片愈伤组织的形成和不定芽分化,而头孢唑林钠在400 mg/L以上时才会对不定芽分化产生不良影响。[结论]为建立矮牵牛的遗传转化体系提供了试验参考。
[Objective] To discuss effects of different antibiotics on regeneration of Petunie hybrida leaf.[Method] With new blade of Petunie hybrida before blossoming as explant,inoculated on the basic culture medium MS+2.0 μg/ml BA+0.2 μg/ml NAA added with different antibiotics(kanamycin,cefazolin sodium and hygromycin),the effects of different antibiotics on leaf regeneration were studied.[Result] 20 mg/L kanamycin,5 mg/L hygromycin could obviously inhibit leaf callus formation and adventitious bud differentiation,while above 400 mg/L,cefazolin sodium could produce harmful effects.[Conclusion] The study will provide reference for establishing genetic transformation system for Petunie hybrida.
