肿瘤相关基因N-myc、Fas、MTA1、nm23-H1对子宫内膜癌细胞增殖转移的影响 被引量:2

摘要 目的探讨原癌基因N-myc、抑癌基因Fas、转移促进基因MTA1及转移抑制基因nm23-H1对子宫内膜癌细胞增殖转移的影响。方法采用实时荧光定量RT-PCR检测正常子宫内膜组织、子宫内膜腺癌未转移患者及转移患者标本各30例中N-myc、Fas、MTA1、nm23-H1的基因表达情况,并行比较分析。结果 (1)子宫内膜癌组织中N-myc、MTA1基因扩增水平均高于正常子宫内膜组织,差异有统计学意义(均P<0.05);(2)子宫内膜癌组织中Fas、nm23-H1基因扩增水平均低于正常子宫内膜组织,差异有统计学意义(均P<0.05);(3)转移子宫内膜癌组织中MTA1基因扩增水平均高于未转移组,nm23-H1基因扩增水平均低于未转移组,差异均有统计学意义(均P<0.05);(4)子宫内膜癌组织转移组与未转移组中N-myc、Fas基因扩增水平均具有差异性,差异有统计学意义(均P<0.05)。结论 N-myc、MTA1基因能促进子宫内膜癌的细胞增殖,Fas、nm23-H1基因能抑制子宫内膜癌的细胞增殖;MTA1基因可促进子宫内膜癌的细胞转移,nm23-H1基因可抑制子宫内膜癌的细胞转移。 Objectlve To study the influence of the original cancer gene N-myc, tumor-suppressor gene Fas, tumor metastasis gene MTA1, metastasis-suppressor gene nm23-H1 to endometrialcarcinoma. Methods By using real time fluorescence quantitative (RT-PCR) detected N-myc, Fas, MTA1, nm23-H1 gene expression in normal endometrium organization 30 cases, endometrial carcinoma cancer (endometrial adenocarcinoma) not-metastasis and metastasis of each group specimens organization 30 cases and the results were analyzed. Resuits (1) The endometrial carcinoma cancer tissue N-myc, MTA1 gene amplification level were higher than normal endometrium tissue, the difference was statistically significant( P 〈 0.05 ) ; (2) Endometrial carcinoma organizations nm23-H1, Fas gene amplification level were lower than normal endometrium tissue, the difference was statistically significant( P 〈 0.05 ) ; ( 3 ) Endometrial carcinoma metastasis group MTA1 gene amplification level were higher than the group not-metastasis, nm23-H1 gene amplification level were lower than non-metastasis group, the difference was statistically significant ( P 〈 0.05 ) ; (4) Endometrial carcinoma group and nonmetastasis group organization N-myc, Fas gene amplification level all had differences, the differences were statistically significant ( P 〈 0.05 ). Conclusions N-myc, MTA1 promote endometrial carcinoma cell proliferation; Fas, nm23-H1 restrain endometrial carcinoma cell proliferation. MTA1 genes promote endometrial carcinoma cell metastasis, gene nm23-H1 restrain endometrial carcinoma cell metastasis.
