华山松无性系种子园遗传多样性分析 被引量:13

摘要 利用ISSR分子标记技术对贵州省平坝华山松无性系种子园及两个不同年份的子代进行遗传多样性研究,结果表明:采用13个ISSR引物对种子园85个无性系进行扩增,共获得了95个标记位点,其中有94个为多态位点,多态位点比率为98.5%。遗传分析表明,种子园无性系有效等位基因数平均值为1.717,遗传多样度(h)平均值为0.402 9,Shannon信息指数(I)为0.585 5,种子园遗传多样性处于较高水平,遗传变异主要存在于种源内;与种子园亲代群体相比,结实初期(1988年)子代群体遗传多样性指数有所降低,结实后期(2007年)子代遗传多样性降低较大,需要采取措施提高种子产量和品质。 An experiment was conducted to study the genetic diversity of clonal seed orchards of Pinus armandii Franch in Ping- ba, Guizhou Province and two offsprings of different years using ISSR markers. Results showed that a total of 95 bands were amplified in 85 clones by 13 primers of ISSR, among which 98.5% of the bands was polymorphie. The effective number of alleles was 1. 717 on average, total genetic diversity was 0. 402 9, and the average of Shannon information index was O. 585 5. A higher diversity was found in the clonal seed orchards. The majority of genetic variation was within prove- nance. The diversity of progeny populations (bearing in 1988 and 2007) were obviously lower than that of the parental populations, especially, the diversity of progeny populations (beating in 2007 ) decreased enormously, indicating that some measures should be taken to improve the genetic quality and seed yield in this orchard.
