摘要
为了研究耶尔森菌强毒力岛(HPI)在银川地区鸡源致病性大肠杆菌中的分布情况,试验根据HPI相关基因irp2和fyuA的参考序列设计引物,采用双重PCR方法对分离的20株鸡源致病性大肠杆菌进行这两种基因的扩增和检测,以及基因克隆和核苷酸序列分析。结果表明:irp2和fyuA基因在鸡源致病性大肠杆菌分离株中的阳性率均为40%(8/20);这两种毒力岛相关基因的核苷酸序列与GenBank中报道的基因核苷酸同源性高达98%以上。说明irp2和fyuA基因具有较高的保守性。
To investigate the distribution of the Yersinia high pathogenicity island (HPI) in pathogenic Escherichia coli strains isolated from chickens in Yinchuan, two pairs of specific primers were designed to amplify fragments of the HPI irp2 and fyuA related genes according to the reference sequences of these two genes,respectively. These two genes were amplified and detected by using the Duplex -PCR in isolated 20 strains of chicken pathogenic Esherichia coli, and then the two genes were cloned and analyzed by nucleotide sequence analysis. The results showed that the positive rates of irp2 and fyuA genes reached 40% (8/20) in the isolated chicken pathogenic Esherichia coll. The results these gene fragments shared more than 98% homology to the reported sequences in GenBank, indicating that the two genes of irp2 and fyuA were highly conservative.
