仔猪大肠杆菌性腹泻的病原检测与免疫防制 被引量:5

Pathogen detection and immune prevention of diarrhea caused by E.coli in piglets

摘要 以江苏姜曲海种猪场的仔猪腹泻病症为研究对象,研究合成了检测肠毒素(ST1、ST2、LT1)、菌毛(K88、K99、987P、F41)以及耶尔森氏菌强毒力岛(high pathogenicity island,HPI)核心基因的特异性引物,用PCR方法对产肠毒素大肠杆菌(enterotoxigenicE.coli,ETEC)和HPI+大肠杆菌进行检测,并对致病性大肠杆菌的免疫防治进行了研究.结果表明:该猪场62.5%的病例存在HPI+大肠杆菌感染,12.5%的病例存在HPI+大肠杆菌及ETEC感染,仅有5%的病例与ETEC感染相关.取分离鉴定的2株大肠杆菌制备灭活抗原,并以氢氧化铝胶为佐剂免疫接种母猪,结果表明,其所产仔猪的腹泻发病率为15.2%,保护率高于仔猪大肠杆菌三价灭活疫苗. Diarrhea caused by E.coli is a common and important disease in piglets,and has an economic significance due to mortality and decreased growth rates.To confirm the contribution of pathogenic E.coli to piglets diarrhea,seven pairs of primers specific to enterotoxin genes,fimbirae genes and high pathogenictiy island(HPI) core gene were synthesized according to the published reference.A total of 40 fecal samples from diarrheic piglets from Jiangsu Jiang Qu Hai boar farm were submitted to PCR rapid detection for enterotoxigenic E.coli(ETEC) and HPI-positive E.coli.The results showed that 25 cases(62.5 %) were infected with HP+E.coli,5 cases(12 %) were co-infected with HPI+E.coli and ETEC,and 2 cases(5 %) were only infected with ETEC.Two E.coli strains were obtained from the fecal samples,and inactivated by formaldehyde followed by culture in TSB and Slanetz broth.When the sows were inoculated with the inactivated bacteria by using Aluminum Hydroxide as adjuvant, the incidence of diarrhea of the piglets were investigated.The data show that only 15.2 % piglets were suffered from diarrhea after immunized with the 2 E.coli strains,which was optimistic than the E.coli trivalent vaccine for newborn piglets from a certain company,and had statistical significant difference compared with normal saline treated group.
