摘要
目的研究双黄花片的HPLC指纹图谱和其主要成分绿原酸与黄芩苷的含量。方法采用HPLC测定,色谱柱为Inertsil ODS-2 C18(250 mm×4.6 mm,5μm);乙腈-0.2%磷酸溶液为流动相,梯度洗脱;检测波长为280 nm;流速为1.0 mL/min;柱温为25℃。结果确定了10个共有峰,10批样品指纹图谱的相似度均在0.8以上,绿原酸和黄芩苷的线性范围分别为:0.256~0.768μg和0.184~0.920μg,加样回收率分别为97.16%和97.23%。结论所建立的HPLC简便、可靠,可用于该制剂的质量控制。
Objective To study the HPLC fingerprints and content of chlorogenie acid and baicalin in Shuanghuanghua tablets. Methods The fingerprints and content were analyzed by HPLC method. The Inertsil ODS-2 C18 column (250 minx4.6 mm,5 μm) was used with acetonitrile -0.2% phosphoric acid solution as mobile phase. Under the condition of gradient elution, the flow rate was 1.0 mL/min, the UV detection wavelength was 280 nm, and the column temperature was 25 ℃. Results Ten marker peaks were separated by HPLC. The similarity factor of fingerprints for 10 batches of samples was over 0.8. The quantitative linear range of chlorogenic acid and baicalin were 0.256-0.768 μg and 0.184-0.920 μg respectively, and the average recoveries were 97.16% and 97.23% respectively. Conclusion The established method in this study on fingerprints of Shuanghuanghua tablets and content determination of chlorogenic acid and baicalin and reliable. It can be used specifically for the quality control of Shuanghuanghua is rapid, simple tablets.
