摘要
建立一个新方法测定多杀菌素发酵液中多杀菌素含量。用分光光度法研究多杀菌素与碘在乙醇中的荷移反应,形成的络合物中多杀菌素与碘分子的摩尔比是2∶3,生成的络合物在290 nm和360 nm处有2个吸收峰,表观摩尔吸光系数分别是3.89×103和2.35×103L/(mol.cm),当多杀菌素浓度为200 mg/L时,相对标准偏差(RSD)为0.712%(n=9)。利用本法测定多杀菌素发酵液中有效成分多杀菌素的含量,并与文献报道的高效液相色谱法进行比较,两者结果基本一致,回收率为98.2%。
To establish a method for rapid determination of the content of Spinosad in fermentation of Sacchar- opolyspora spinosa. The charge transfer reaction between Spinosad and iodine in aleoholmedium was investigated by speetrophotometry. The apparent molar absorptivity of complex was 3.89 ×103L/(mol · cm) at 290nm and 2.35× 103 L/( tool ·cm)at 360 nm. The composition ratio and stability constant of charge transfer complex were 2:3 re- spectively. The average standard deviation of nine determinations was 0. 712% when the concentration of Spinosad was 200 mg/L. The proposed method has been applied to the determination of Spinosad in fermentation of Sacchar- opolyspom spinosa with satisfactory results.
