摘要
研究了一种快速测定普洱茶叶及其提取物中茶多糖含量的方法:先对普洱茶叶进行水提(或将普洱茶粉直接溶于水),再进行80%醇沉,然后用0.5 mol/L的盐酸溶液在40℃下水解30 min,用聚酰胺层析柱分离洗脱,最后用苯酚-硫酸法测定茶多糖的含量。该方法可有效去除普洱茶中茶色素等物质的干扰,准确测定出水溶性茶多糖的含量。
A method was developed for the rapid determination of water - soluble tea polysaccharide content in Pu' er tea and its extract. Firstly, Pu' er tea leaves were extracted through water, or Pu' er tea powder was directly dissolved into water; secondly, the extraction solution was sedimentated with 80% ethanol, and then they were hydrolyzed by hydrochloric acid solution (0.5 mol/L) for 30 min at 40 ℃ ; thirdly, they were isolated and eluted through polyamide chromatographic column; finally, the water - soluble tea polysaccharide content was determined by using phenol - sulfuric acid method. The results indicated that this method could effectively wipe off the interference from tea pigment etc. in Pu' er tea, and accurately determine the content of water - soluble tea polysaccha- ride.
