摘要
用离子色谱法建立一种简单、快速、准确、灵敏的半乳糖醛酸和葡萄糖醛酸的测定方法.多糖水解液经过简单的中和、稀释、过滤后直接进样分析.半乳糖醛酸和葡萄糖醛酸具有较高的分离度(R=4.2);两者在20~200 mg/L质量浓度范围内都具有良好的线性,其相关系数均为r=0.999 0;重复性和回收率都满足了仪器分析要求;两种化合物的检测限分别为100 ng和50 ng.
A simple, fast, accurate and highly sensitive method for the determination of galacturonic acid and glucuronic acid in the hydrolyzed polysaccharide solution by Ion Chromatography-PAD was established. After neutralization, dilution, and filtration, the hydrolyzed polysaccharide solution was injected directly. Galacturonic acid and glucuronic acid exhibited good resolution (R=4.2). Two linear regression equations of y=27 469x+136 911(r =0. 999 0)and y=66 379x+347 163( r=0. 999 0)were obtained for galacturonic and glucuronic acids, respectively. The linear ranges of the two compounds were 20 ~ 200 mg/L. The good accuracy and recovery rate met the demands of the instrument analysis. The limit of detection was 0. 1μg and 0. 05μg, respectively.
