壮肾1号颗粒对阿霉素肾病大鼠蛋白尿模型的外周血CD_(44)表达的影响 被引量:2

Experimental Research on Zhuangshen Yihao Granule on the Expression of CD44 in Peripheral Blood of ADR Nephrosis Rats

摘要 目的:观察壮肾1号颗粒对阿霉素肾病大鼠蛋白尿模型的外周血CD44、尿β2-MG的影响。方法:用尾静脉一次性注射阿霉素制备肾炎蛋白尿模型,分为5组,分别为空白对照组、模型对照组、洛汀新组、壮肾1号颗粒低剂量组、壮肾1号颗粒高剂量组,观察8周,检测大鼠的尿24h尿蛋白定量、β2-MG,取血流式细胞术检测CD44阳性细胞数。结果:实验8周末各治疗组(壮肾1号颗粒高、低剂量组及洛汀新组)阿霉素肾病大鼠24h尿蛋白定量下降,以壮肾1号颗粒高剂量组最为明显(P<0.05);尿β2-MG的含量降低,以壮肾1号颗粒高剂量组最为明显(P<0.05);阿霉素肾病大鼠CD44阳性细胞数降低,以壮肾1号颗粒高剂量组最为明显(P<0.05)。结论:壮肾1号颗粒能够在一定程度上减少阿霉素肾病大鼠尿蛋白的排泄量,对肾小球有保护作用;减少阿霉素肾病大鼠尿β2-MG的含量,对肾小管有保护作用;降低CD44阳性细胞数降低,提示壮肾1号颗粒通过对阿霉素肾病大鼠外周血CD44阳性细胞数的降低,减轻炎症性损害的发生,影响机体致炎及免疫损伤,从而减少尿蛋白的可能机制。高剂量组对于上述作用较中药低剂量组、洛汀新组疗效较好。 Objective:To observe the effect of Zhuangshen Yihao Granule on CD44 expression in peripheral blood and urine β2 microglobulin(β2-MG)of the proteinuria model of nephrosis rats induced by Adriamycin(ADR Nephrosis Rats).Methods:The proteinuria models of nephrosis rats were established by injecting ADR into the tail vein.And then they were divided into groups randomly as follows:the model group,the Lotensin group,the low-dose group and the high-dose group of Zhuangshen Yihao Granule.Certainly,they were observed with the normal group.Before the experiment and in the 8th week of the experiment,we tested 24 hours urinary protein,urine β2-MG and the number of CD44 positive cells in peripheral blood by flow cytometry.Results:In the 8th week,except the normal group and the model group,the levels of 24 hours urinary protein,urine β2-MG and CD44 positive cells of all groups were uniformly reduced,especially in the high-dose group of Zhuangshen Yihao Granule(P0.05).Conclusion:Zhuangshen Yihao Granule can reduce the levels of 24 hours urinary protein,urine β2-MG and CD44 positive cells to a certain extent.That is to say,it can not only exert a protective effect on glomerulus and renal tubule,but aslo reveal that it may abate the impairment of the inflammation,impact on proinflammatory cytokines and immune injury to decreased urine protein excretion accordingly.Furthermore,the above curative effects of Zhuangshen Yihao Granule in the high-dose group are better than those in the low-dose group and the Lotensin group.
