摘要
以 N-羟基琥珀酰亚胺 - α-萘乙酸酯 (SINA)为氨基酸的柱前衍生试剂 ,反相高效液相色谱分离测定了 1 5种氨基酸。采用含 1 0 mmol/ L p H 5.0的乙酸 -乙酸钠缓冲溶液的甲醇 -乙酸乙酯 -水 (1 0 / 2 / 88,V/ V/ V)溶液为流动相体系 ,分离测定了 7种氨基酸 ;用甲醇 -乙酸乙酯 -水 (2 6/ 2 / 77,V/ V/ V)分离测定了 5种氨基酸 ;用甲醇 -乙酸乙酯 -水 (45/ 2 / 53,V/ V/ V)分离测定了 3种氨基酸。检测限均达到 pmol级 。
NsuccinimidylαNaphthylacetate (SINA) was used as a precolumn derivatizing reagent to separate and determine 15 amino acids. With a mobile phase of methanol ethyl acetate water (10/2/88, V/V/V) containing 10 mmol/L of pH 5.0 acetate buffer, Trp, Asp, Glu, Ser, Gly, Thr and Ala were separated. After the methanol content in mobile phase was increased to 26%, CySH, His, Tyr, Met and Val were separated. When methanol content in mobile phase was 45%, Ile, Phe and Lys were separated. The detection limits were at pmol range(S/N=3) with good reproducibility.
