血管紧张素转换酶基因多态性与原发性高血压的关系 被引量:2

Relationship between Angiotensin Coverting Enzyme Gene Polymorphism and Essential Hypertension

摘要 采用多聚酶链反应 ( PCR)技术检测了 154例汉族男性原发性高血压患者 ( EH组 ) ACE基因第 16内含子中 2 87bp DNA片段的插入 /缺失 ( I/ D)多态性 ,并与 183例健康查体者 (对照组 )进行比较。结果显示 ,两组年龄、甘油三酯 ( TG)、胆固醇 ( CH)、高密度脂蛋白胆固醇 ( HDL - C)、Apo A1、Apo B和血糖 ( Glu)均无显著性差异。对照组 ACE基因 I/ D多态 DD、 D、 基因型频率分别为 0 .2 2 4、0 .4 86、0 .2 90 ,其 D和 等位基因频率分别为 0 .4 67和 0 .533。 I/ D多态分布符合 Hardy- Weinberg法则 ,达到遗传平衡 ,具有群体代表性。EH组中无 EH家族史 ( 97例 )和有家族史者 ( 57例 )的 DD、 D、 基因型频率分别为 0 .351、0 .4 4 3、0 .2 0 6和 0 .4 0 4、0 .4 38、0 .158,D和 等位基因频率分别为 0 .572、0 .4 2 8和0 .62 3、0 .377。EH组无和有家族史者基因型 DD和等位基因 D均显著高于对照组 (分别为 P<0 .0 2 5,P<0 .0 2 5;P<0 .0 0 1,P<0 .0 0 5) ,但两组之间基因型和等位基因分布无显著性差异。认为汉族男性 EH的发生与 ACE基因 I/ D多态性相关联 ,基因型 DD和等位基因 D可能是 The 287bp insertion/deletion(I/D)polymorphism in intron 16 of angiotensin coverting enzyme(ACE)gene was examined by polymerase chain reaction(PCR)in 154 Chinese Han male patients with essential hypertension(EH),and compared with 183 controlsThe result showed that no significant differences were observed in age,TG,Ch,HDLC,Apo Al,ApoB and glucose between two groupsThe frequencies of ACE genotypes DD,Ⅰ[KG-3]D and II were 0224,0486,0290 respectively and the frequencies of D and I alleles were 0467,0533 respectively in 183 controlsThe distributions of I/D polymorphism of ACE gene were in agreement with Hardy-Weinberg equilibriumThe frequencies of ACE genotypes DD,ID and Ⅱ were 0.351,0.443,0.206 repectively and the frequencies of D and I alleles were 0.572,0.428 respectively in 97 EH.The frequencies of ACE gene types DD,ID and II were 0404,0438,0158 respectively and the frequencies of D and I alleles were 0623,0377 respectively in 57EH with family historyThe frequencies of genotype DD and allele D were significant higher in EH and EH with family history compared with controls (P<0025,P<0025;P<0001,P<0005 respectively),but no significant differences were observed in genotype DD and allele D between EH and EH with family historyThis suggest that the I/D polymorphisms of ACE gene are relative to EH and genotype DD and allele D may be the fact of genetic predisposition of EH in Chinese Han male people
