胰头夹闭法致大鼠急性坏死性胰腺炎动物模型的建立 被引量:6

Study on establishment of an ischemic and necrotizing pancreatitis model by obstructing head of pancreas in rats

摘要 目的通过暂时夹闭胰头部的方法建立一种新型大鼠急性坏死性胰腺炎(ANP)的动物模型。方法将健康雄性SD大鼠180只随机分为:对照组、假手术组、ANP模型组,每组60只。ANP模型组开腹后钝性分离胰腺周围韧带直至胰头部,无创血管钳夹闭胰头2 h后松开。在造模完成后分别于0 h、4 h、12 h、24 h、48 h、72 h各时间点随机取10只大鼠检测血淀粉酶、脂肪酶,取胰腺组织标本检测湿/干重比,行病理组织学观察。结果 ANP模型组大鼠血淀粉酶、血清脂肪酶与对照组和假手术组比较,差异显著(P﹤0.05)。病理组织学观察可见造模后随时间进程胰腺炎呈加重表现(P<0.05)。结论通过夹闭胰头部的方法建立了一种新型大鼠急性坏死性胰腺炎动物模型,为急性坏死性胰腺炎的发病机制及药物干预研究提供了一种较为理想的动物模型。 Objective To establish an acute ischemic and necrotized pancreatitis model by obstructing head of pancreas in rats. Methods A total of 180 rats were randomly divided into control group (n = 60) , sham - operation group (n = 60 ) and acute necrotizing pancreatitis (ANP) model group (n = 60). The pancreas and pancreatic tail -segment was dissociated and the head of pancreas was occluded in rats to form model of acute necrotizing pancreatitis, blood vessel forceps Was loosed after two hours. All rats were killed at 0 h, 4 h, 12 h, 24 h ,48 h and 72 h after modeling. The level of serum amylase and lipase, wet to dry weight ratio of pancreas and histopathological scores for pancreas were studied. Results The levels of serum amylase [0 h(1 401.7 ±16.5), 4h(1 461.2 ±41.0), 12 h(2 620.3 ±226.3), 24 h(3 905.2 ±2 20.6), 48 h (3 080.0 ±106.2)and 72 h(2 816.3 ± 141.5 )] and serum levels of lipase [ at 0 h (364.0 ± 12.8 ) ,4 h (382.4 ± 15.5 ), 12 h (426.9 ± 12. 7 ) ,24 h (479.0 ± 12.9 ) ,48 h (507.5 ± 14.1 )and 72 h (500.7 ± 16.6) ] in group of ANP niodel were significantly increased after establishment of the model. The tissue damage of pancreas became more serious after establishment of the model. Conclusion The model shows typical ischemic and necrotizing pancreatitis in rats by obstructing head of pancreas. It is an ideal and clinically relevant animal model for study of nosogenesis and therapeutical effect of pancreatitis.
