胚胎脊髓移植联合应用神经生长因子及尼莫地平对成鼠损伤脊髓的影响 被引量:3

Effect of combining use of fetal spinal cord grafts,NGF and Nimodipine on the injured spinal cord of adult rats

摘要 目的 :研究胚胎脊髓移植联合应用神经生长因子 (NGF)及尼莫地平 (ND)对损伤脊髓的修复作用。方法 :90只Wistar大鼠半切洞脊髓损伤后 ,随机分为 3组 :(1)单纯胚胎脊髓移植组 (A组 ) ,(2 )移植 +NGF组 (B组 ) ,(3)移植 +NGF +ND组 (C组 )。观察组织形态学、运动诱发电位及行为学变化。结果 :C组的运动诱发电位潜伏期在早期比A组、B组皆有明显缩短 (P <0 0 5 )。以后肢运动功能的Tarlov评分看 ,B组和C组的后肢运动功能明显优于A组 (P <0 0 5 )。病理观察显示 ,移植的胚胎脊髓可在宿主内分化、发育 ,并可挽救损伤神经元 ,同时可与宿主脊髓结构连接。结论 :胚胎脊髓移植与NGF、ND合用 ,可能是脊髓损伤的一种有效疗法。 Objective:To study the effect of fetal spinal cord grafts,Nerve growth factor(NGF) and Nimodipine(ND) on repairing injured spinal cord.Method:90 rats were hemisection cavity injured at the spinal cord,then divided into transplantation group(A),Transplantation+NGF treated group(B),Transplantation+NGF+ND treated group(C).Motor evoked potentials(MEP),motor function of hindlimbs(Tarlov′s Score) and pathological changes were observed.Results:Tarlov′s score in B and C groups were higher than that in A group(P<0 05),NGF and ND could shorten the latencies of MEP.The pathological observation indicated that fetal spinal cord(FSC) could grow and differentiate in the host spinal cord and rescue the injured neurons,and the fiber connections between FSC and host spinal cord were reconstructed.Conclusions:The combined application of FSC transplantation,NGF and ND may be an effective therapy for spinal cord injury.
