摘要
目的:探讨姜黄素诱导人肺癌细胞凋亡的分子机制。方法:MTT法检测姜黄素对肺癌细胞活力的影响,流式细胞计数法检测姜黄素对肺癌细胞凋亡及线粒体膜电位的影响,Caspase活性试剂盒检测姜黄素处理后肺癌细胞Caspase活性的变化,Western-blot检测姜黄素对胞质中细胞色素C的含量及肺癌细胞Akt信号通路的活性的影响。结果:20μmol/L以上浓度的姜黄素作用48h后可显著抑制A549细胞的活力,显著诱导细胞发生凋亡,姜黄素作用24h后肺癌细胞中的线粒体跨膜电位明显下降,细胞色素C释放到胞质增多,Caspase-9、-3活性显著增强,磷酸化Akt水平显著下降。结论:抑制Akt信号通路活性诱发线粒体通路介导的细胞凋亡可能是姜黄素抑制肺癌细胞的重要机制。
Objective: To investigate the molecular mechanism of curcumin induced apoptosis in human lung cancer cell.Methods: Cell viability was determined by MTT assay,the apoptosis and mitochondrial membrane potential were detected by flow cytometry,caspase activity was assayed,and cytoplasmic cytochrome C and Akt signaling pathway activity was assayed by Western-blot in lung cancer cells.Results: Twenty μmol/L or more concentrations of curcumin treatment on A549 cells for 48 hours significantly inhibited the cell activity,significantly induced apoptosis;curcumin treatment for 24 hours significantly decreased mitochondrial membrane potential,cytochrome C secretion increased,caspase-9 and-3 activity was significantly enhanced,while phosphorylation of Akt was significantly decreased.Conclusion: Inhibition of Akt signaling pathway activity and eventually induced mitochondrial-mediated apoptosis may be an important mechanism of curcumin-induced apoptosis in lung cancer cells.
