柚皮素对K562细胞的抑制作用与Caspase酶活性、PCNA蛋白表达的关系 被引量:2

摘要 目的本研究旨在探讨柚皮素对K562细胞的生长抑制作用与Caspase酶的活性及细胞增殖核抗原(PCNA)蛋白表达的关系。方法用50mol/L,200mol/L的柚皮素分别作用K562细胞后,Caspase分光光度计法检测细胞内Caspase-3和Csapase-9酶活性的改变;用细胞免疫化学的方法检测PCNA蛋白的表达。结果在柚皮素作用下,K562细胞内Caspase-3和Caspase-9活性增强(P<0.05),PCNA的表达显著降低(P<0.05)。结论柚皮素对K562细胞的抑制作用可能是通过细胞增值抑制和上调Caspase-9和Csapase-3活性诱导细胞凋亡而实现。 Objective To investigate the relation ofapoptosis ofK562 cells induced by naringenin to expression of PCNA and enzyme activity of Caspase. Methods The activity of Caspase-3 and Caspase-9 in K562 cell line induced by naringenin was detected by Caspase Colorimetric Assay. PCNA protein ofK562 cell line treat- ed by naringenin was detected by immunhistochemical staining. Results The activity of Caspase-3 and Caspase-9 increased remarkably with treatment ofnaringenin, while PCNA protein in treatment group decreased remarkably than that in control group. Conclusion The activity of Caspase-3 and Caspase-9 and cell cycle arrest may contribute to the apoptosis of K562 cell in vitro induced by naringenin.
