摘要
建立血浆中头孢曲松反相 HPL C测定法 ,用于胸外术后患者头孢曲松血药浓度的测定。血浆样品经乙腈沉淀蛋白等预处理后 ,于 ODS柱上 ,以甲醇 - 0 .1mol/ L 乙酸铵缓冲液 (2 0∶ 80 )为流动相进行分离测定。线性范围 0 .2～ 80 mg/ L,沉淀蛋白回收率 91.6 %～ 94.4% ,加样回收率 10 1.5 %～ 10 3.2 % ,日内 RSD1.0 9%～ 2 .6 5 % ,日间 RSD 4.42 %～ 5 .13%。本法准确可靠 ,用于胸外术后患者头孢曲松血药浓度测定取得良好结果。
A simple HPLC method was established for the determination of ceftriaxone in human plasma. Acetonitrile was used as deproteinizing agent for the plasma samples. The chromatographic separation was on ODS column with a mobile phase consisting of methanol and 0.1mol/L ammonium acetate buffer (20∶80). The calibration curve was linear within 0.2~80mg/L. The deprotein recoveries of ceftriaxone were 91.6% ~94.4%. The recoveries of methodology were 101.5%~103.2%. Inter day and intra day RSD ranged from 1.09% ~2.65% and 4.42%~5.13% respectively. This method is accurate and reliable. It has been applied successfully to the determination of ceftriaxone in patients after operation on chest.
