固相萃取-高效液相色谱法快速测定血中6种苯二氮类药物浓度及其临床应用 被引量:4

Simultaneous determination of six benzodiazepines drugs in serum by HPLC with solid phase extraction

摘要 目的 建立SPE HPLC法同时测定血清中 6种苯二氮类药物 (地西泮、硝西泮、氯硝西泮、三唑仑、艾司唑仑、阿普唑仑 )浓度。方法 用固相萃取技术处理含药血清 ,色谱柱为Nova pakC1 8柱 ,3 9mm× 15 0mm ,4 μm ;流动相为 0 0 1mol L磷酸盐缓冲液 (pH 2 15 )∶乙腈 (71∶2 9) ;检测波长 2 2 3nm ;流量 1ml min ,柱温 30℃。结果  6组分分离良好 ,在 0 0 1~ 10 0 0 μg ml峰面积与浓度呈良好的线性关系(r>0 999) ,相对回收率为 96 %~ 10 4 % ,日内、日间RSD均小于 9%。结论 该色谱条件可同时测定血清中此类药物 ,具有快速、简便、准确可行等特点 ,非常适用于此类药物中毒患者的快速鉴定 。 Objective To establish a rapid and simple analysis method for the determination of 6 benzodiazepines drugs (diazepam, nitrazepam, clonazepam, triazolam, estazolam, alprazolam) in serum simultaneously by HPLC with solid phase extraction.Methods Drugs in serum were extracted with accubond SPE C 18 column. The analytical column was Nova-pak C 18 column (3.9 mm×150 mm,4μm). The mobile phase consisted of 0.01 mol/L phosphate buffer solution (pH 2.15)-acetonitrile(71∶29,V/V).The detection wavelength was 223 nm and the flow rate was 1 mL/min, column temperature was 30℃.Results Six drugs were extracted completely, impurities were removed thorough. Six drugs were separated well under the chromatographic condition. Every linear range of the six drugs was 0.01-10 μg/mL and all correlation coefficient was more than 0.9990. The relative recoveries of the 6 drugs was 96% to 104% respectively. The relative standard deviation(RSD) of within day and between day were all less than 9%.Conclusions This method is found to be rapid, simple,accurate, sensitive and more suitable for identifying the benzodiazepines drugs toxicosis and therapeutic drug monitoring application.
