摘要
目的 :建立高效液相色谱法测定血浆中加替沙星浓度。方法 :血浆用二氯甲烷提取 ,色谱柱为ZorbaxC18(4.6mm×2 5 0mm) ,流动相为 0 .0 1mol·L-1磷酸二氢钾 甲醇 三乙胺 (6 5∶34.8∶0 .2 ) ,检测波长 2 92nm ,流速 1.0ml·min-1,以司帕沙星为内标。结果 :加替沙星浓度线性范围 0 .0 1~ 5 μg·ml-1,最低检测浓度 0 .0 1μg·ml-1,绝对回收率为 85 .1% ,相对回收率为10 0 .6 %。日内 ,日间RSD分别为 3.0 %和 3.7%。结论 :该方法适用于人体药物动力学研究。
Objective: To determine the gatifloxocin concentration in human plasma by HPLC.Method: Gatifloxacin was extracted from plasma with dichloromethane. It was chromatographed on an Zorbax ODS column with spafloxacin as internal standard. The mobile phase consisted of 0.01 mol·L -1 KH 2PO 4 methanol triehylamine (65∶ 34.8 ∶ 0.2 ) adjusted to pH 3.0 with orthophosphoric acid. The flow rate was 1.0 ml·min -1 and the detecting wavelength was at 292 nm.Result: The calibration curve was linear from 0.01 to 5.0 μg·ml -1 . The detecting limit of gatifloxacin was 0.01 μg·ml -1 . The absolute and relative recoveries of gatifloxacin in plasma were 85.1% and 100.6% , respectively. RSD within day and between day were 3.0% and 3.7% , respectively.Conclusion: The method can be used to investigate the pharmacokinetics of gatifloxacin in human plasma.
