摘要
目的 研究碱性成纤维细胞生长因子 (basic fibroblast growth factor,b FGF)在体外对人晶状体上皮细胞增殖的影响及机制探讨。方法 取人晶状体前囊膜培养 ,加入不同浓度b FGF(1μg· L- 1、10 μg·L- 1、10 0 μg·L- 1 ) ,48h后免疫组化测增殖核细胞抗原阳性面积率。取人晶状体前囊膜免疫组化测 b FGF受体。结果 一定浓度 (1~ 10 0 ug· L- 1 )的 b FGF在体外有促人晶状体上皮细胞增殖的作用 ,且呈剂量相关性。人晶状体上皮细胞 b FGF受体表达阳性。结论 b FGF通过受体介导可促人晶状体上皮细胞增殖 ,在维持人晶状体上皮细胞的正常生理和后发障的发病机制中占有重要的地位。
Objective To assess the effect of basic fibroblast growth factor(bFGF) on the mitosis of human lens epithelial cells and discuss its mechanism.Methods Human lens anterior capsules were cultured in vitro ,treeted with different concentration of bFGF(1μg·L -1 、10μg·L -1 、100μg·L -1 )for 48 hours,then the positive area ratio of prolifterating cell nuclear antigen (PCNA) and the existence of receptors for bFGF in immunohistochemistry were estimated.Results bFGF(1~100μg·L -1 )increased the mitosis of human lens epithelial cells and the existence of receptors for bFGF in human lens epithelial cells was confirmed.Conclusion bFGF plays an important role in the physiological condition of human lens epithelial cells and the formation of the after cataract.
