摘要
目的 进一步探讨Alzheimer病 (AD)与载脂蛋白E(ApoE)的关联性。 方法 通过对济南铁路地区 2 0 14名 60岁以上的老年人进行痴呆流行病学调查 ,随机选出AD患者 3 6例、血管性痴呆患者 (VD) 2 7例和非痴呆老人 (ND) 3 6例 ,取外周血抗凝 ,酚、氯仿 /异戊醇法提取DNA ,应用聚合酶链反应 限制性片段长度多态性分析(PCR RFLP)技术对ApoE基因的多态性进行分析。 结果 AD、VD患者与ND的ApoEε4、ε3、ε2的等位基因频率依次为 0 2 5 0、0 681、0 0 69和 0 0 74、0 83 3、0 0 93及 0 0 5 6、0 889、0 0 5 6。AD患者的ApoEε4基因频率显著高于对照组 (P <0 0 1) ,ApoEε3基因频率明显低于对照组 (P <0 0 5 )。不含、含 1个、含 2个ApoEε4 AD的患病率分别为 2 7 6%、60 0 %、10 0 %。随ApoEε4基因剂量的递增AD的发病年龄提前。 结论 ApoEε4在AD中增高具有较高的特异性 。
Objective To discuss the association of Alzheimer’s disease(AD)and apoE. Methods By the epidemiology on 2 014 cases of the elderly beyond 60 years old in Jinan Railway area, 36 cases of AD,27 cases of VD and 36 cases of ND were screened out. DNA was extracted from the peripheral blood samples with the phenolchloroform procedure. ApoE genetypes were investigated with the methods of polymerase chain reaction PCR and restriction fragment length polymorphism(RFLP). Results The frequencies of ApoE ε4,ε3 and ε2 was 0 250,0 681 and 0 069 in AD, 0 074,0 833 and 0 093 in VD, as well as 0 056,0 889 and 0 056 in ND. The frequency of ApoE ε4 was significantly higher in AD than in ND ( P <0 01),The frequency of ApoE ε3 was significantly lower in AD ( P <0 05). The morbidity rate in AD with none ε4,1 and 2 was 27 6%,60 0% and 100%. Following the increase of dose of ApoE ε4 in AD, it occurs earlier. Conclusions There was a significant association with the increase of ApoE ε4 in AD. ApoE ε4/4 plays an important role in the diagnosis of AD.
