摘要
为探讨白细胞介素 -6( IL -6)在卵巢切除诱导的大鼠骨吸收中的作用及补肾中药密骨片对其的影响。将 3 0只 SD大鼠随机分为假手术组 ( Sham ) ,卵巢切除组 ( OV X)及密骨片治疗组 ( OVX-MGP)。分离三组大鼠右股骨骨髓上清 ,以 10 %~ 60 %浓度加入 45Ca标记的胎鼠颅骨培养液中。观察骨吸收活性并通过 EL ISA测定骨髓上清 IL-6含量。结果示骨髓上清 IL-6浓度与骨吸收活性呈量效关系 ;OVX组骨吸收活性较 Sham组显著升高 ,OVX-MGP组较 OVX组明显下降 ( P均 <0 .0 1) ;OVX组骨髓上清 IL-6浓度是 Sham组的 2倍 ,与 OV X比较 ,O VX-MGP组 IL -6含量下降 5 2 % ( P均 <0 .0 1)。表明卵巢切除导致内源性骨吸收因子 IL -6产生增加 ,密骨片抗骨吸收作用部分是通过降低 IL-6产生而介导的。
In order to study the role of endogenous interleukin 6 (IL 6) in the induced bone resorption in ovariectomized rats and the effect of “MIGU PIAN” (MGP, a TCD preparation for reinforcing the kidneys) on IL 6, thirty SD rats were randomly divided into three groups: sham surgery (SS, or non ovariectomized), ovariectomized (OV), and ovariectomized and MGP treated (OVMGP). Bone marrow supernatants were separated from the rat right femora of the three groups and added at 10 % to 60 % concentration to the culture media of 45 Ca labelled fetal rat cranial bones. Then the bone resorption activities (BRA) were examined and IL 6 levels in the supernatants were measured by ELISA (enzyme linked immunosorbent assay). The results showed: (1) dose effect relationship between IL 6 concentration and BRA; (2) much raised BRA of OV group in comparison with that of SS group and much lowered BRA of OVMGP group in comparison with that of OV group (P < 0.01); (3) IL 6 level of OV group being two times of that of SS group and 52 % drop of IL 6 level of OVMGP group in comparison with that of OV group (P < 0.01). It was concluded that ovariectomy results in endogenous BRA factor IL 6 increase and BRA resisting effect of MGP is obtained partially through reducing IL 6 production.
