摘要
为优化玉米超氧化物歧化酶(SOD)提取与纯化工艺。研究不同浸提条件及沉淀分离工艺对玉米SOD提取及纯化效果的影响。最佳诱导和浸提的原料与缓冲液配比分别为1∶1.5~1∶3、1∶2,最佳诱导和浸提时间分别为25 h^30 h、1.5 h。采用50、60℃分别处理15、10 min两次热变性除杂蛋白、丙酮沉淀SOD的纯化效果最好。DEAE-纤维素柱层析纯化后,SOD比活为1 699.7 U/mg,回收率为31%,纯化倍数为30.7。该方法高效且低毒污染少,适于工业化生产。
To optimize the extraction and purification technology of superoxide dismutase from corn.Effect of different technique on preparation of corn SOD was studied.The optimum inducement and digestion condition is material and the buffer solution ratio was 1∶1.5-1∶3,1∶2 respectively,the optimum time was 25 h-30 h,1.5 h respectively.The optimum purification technique was 50 ℃,60 ℃ treatment for15 min,10 min respectively,and SOD was deposited by acetone.Carried on the DEAE-cellulose column chromatographic analysis purification to extraction SOD,SOD had a specific activity of 1 699.7 U/mg.The yield rate was 31 %,and themultiplication was 30.7 times.The SDS-PAGE electrophoresis result had indicated that the purification SOD achieved the electrophoresis to be pure.
