四种邻苯三酚自氧化法测定超氧化物歧化酶活性方法的比较 被引量:92

摘要 邻苯三酚自氧化法(420nm法)与邻苯三酚/DTT终止法测定SOD酶活力,结果基本一致,其转换系数(CC)为1.03;改进的邻苯三酚自氧化法(325nm法)、微量邻苯三酚/Vc终止法测定SOD酶活力与420nm法差别较大,其转换系数分别为2.51、5.51.在测定SOD酶活力时,可根据不同的测定方法采用转换系数进行校正;实验表明:终止剂维生素C(Vc)和二硫苏糖醇(DTT)对SOD酶活力的测定无影响;325nm法与Vc法灵敏度较高,可用于SOD活力较低的样品. There is no distinction in determining SOD activity between pyorgallol autoxidation(420nm) and pyorgallol autoxidation/dithiothreitol(DTT) method.The conversion coefficient is 1.03;The results are different in determining SOD activity with the method of improved pyorgallol autoxidation (325nm) and trifling pyorgallol autoxidation/vitaminC(Vc) as compared with pyorgallol autoxidation(420nm).And its conversion coefficient is 2.51 and 5.51 respectively.They can be conversed with conversion coefficients in different methods of determining SOD activity. The tests also show there are no effects in determining SOD activity using Vc and DTT. The improved pyorgallol autoxidation(325nm) and trifling pyorgallol autoxidation/Vc can be used in determining lower SOD activity sample due to their high sensitivity.
