摘要
目的 :研究骨巨细胞瘤微血管生成能力及意义 ,并探讨各种细胞的不同来源和功能。方法 :用LSAB法检测骨巨细胞瘤病理标本微血管密度 (MVD)和特征及TGFβ1和TGFβ3 表达。结果 :病理分级之间MVD :Ⅲ级 >Ⅱ级 (P <0 .0 5 ) ,Ⅲ级 >Ⅰ级 (P <0 .0 1) ,瘤组织分化越差 ,微血管异质性越强。TGFβ1表达与MVD增加显著相关 (P <0 .0 1) ,与术后复发等不良预后相关 (P <0 .0 5 )。TGFβ3 表达与病理分级相关 ,而与微血管密度无关 (P <0 .0 5 ) ,与外科分期有关 (P <0 .0 5 ) ,与术后复发等不良预后有关 (P <0 .0 5 )。TGFβ1与TGFβ3 阳性表达相关 (P <0 .0 5 ) ,其表达率之间有显著差异 (P <0 .0 1)。结论 :MVD、TGFβ1和TGFβ3 阳性表达可作为骨巨细胞瘤不良预后的指标。FC可能是真正瘤细胞 ,MGC和MC可能是瘤细胞分泌细胞因子趋化而来的破骨细胞和前体破骨细胞。
Objective:To investigate tumor angiogenesis and to determine the origin and function of each kind of cells in GCT. Methods: LSAB method was developed to detect the MVD and its feature, and the expression of TGFβ 3 and TGFβ 3 in GCT. Results: The MVD significantly correlated with pathological grade and the poor prognosis( P <0.05), But it did not correlated with the location and surgical stages. TGFβ 1 expressed in cytoplasm of MGC, MC, FC and EC in GCT. No significant correlation was found between the TGFβ 1 expression and pathological grade. The positive expression of TGFβ 1 significantly correlated with the high MVD( P <0.01) and poor prognosis( P <0.05), but it did not correlate with surgical stage( P >0.05). TGFβ 3 expressed in cytoplasm of MGC and MC. The positive expression of TGFβ 3 significantly correlated with the surgical stage, and lower malignance( P <0.05) and poor prognosis( P <0.05). But no correlation with the MVD. The expression of TGFβ 1 (24.7% ) and TGFβ 3(55.6%) was significantly different from and correlated with each other( P <0.05). Conclusions: MVD increasing of MVD and the expression of TGFβ 1 and TGFβ 3 maybe regarded as useful prognostic markers of GCT. FC maybe tumor cell. MGC and MC maybe the osteoclast and osteoclast precurser migrated by cytokines secreted by FC. But this hypothesis needs to be studied further.
