摘要
目的 探讨大肠癌、大肠腺瘤及正常粘膜中nm 2 3 H 1,p16蛋白的表达。方法 应用免疫组化ABC法对 3 0例大肠癌、2 5例大肠腺瘤及 2 5例正常大肠粘膜中nm 2 3 H 1、p16蛋白的表达进行测定。 结果 大肠癌组织中nm 2 3 H 1与 p16阳性表达率分别为 66.7%和 40 .0 % ;p16在大肠癌中的表达水平明显低于正常大肠粘膜及腺瘤中的表达 (P <0 .0 5 ) ,而nm 2 3 H 1表达在各组间虽呈下降趋势 ,但无显著性差异 (P >0 .0 5 ) ;两者在大肠癌中的表达存在相关关系 (P <0 .0 5 ) ,并与癌组织的分化程度、有无淋巴结转移及患者的生存时间等均有明显相关性 (P <0 .0 5 )。结论 抑癌基因 p16的失活、nm 2 3 H 1基因的低表达可能与大肠癌的发生、发展、转移及预后有关。
Objective To investigate the expression of nm23 H1 and p16 in colorectal carcinoma,adenoma and normal mucosa.Methods The expression of nm23 H1 and p16 in 30 cases of colorectal carcinoma,25 adenoma and 25 normal mucosa were determined by ABC immunohistochemical technique.Results Positive expression rates of nm23 H1 and p16 were 66.7%,40.0% in colorectal carcinoma.The expression level of p16 in cancerous tissue was lower than that in normal mucosa and adenoma( P <0.05).No significant difference in the expression of nm23 H1 was found in different colorectal tissues( P >0.05).The positive rates of nm23 H1 and p16 expression in different pathological grading,lymph node metastasis and post operative survival(3 years) were significantly different( P <0.05).There was significant correlation between the expression of nm23 H1 and p16( P <0.05).Conclusion The deletion of p16 and nm23 H1 low expression might play an important role in the carcinogenesis,development,metastasis and prognosis of human colorectal carcinoma.
